ATTIVITÀ SCIENTIFICA
La Dott.ssa Rufini svolge la sua attività di ricerca nel Laboratorio di Trasduzione del Segnale (www.labst.org), Unità di Malattie Rare, presso il Dipartimento di Biomedicina e Prevenzione dell’Università di Roma “Tor Vergata”. Durante il dottorato, il lavoro della Dott.ssa Rufini è stato incentrato sullo studio del cross-talk tra caspasi e tirosin-chinasi nel controllo del pathway di apoptosi. Da più di 15 anni la sua attività di ricerca è focalizzata sullo studio della Atassia di Friedreich, una malattia neurodegenerativa rara, per la quale non esiste cura. Attualmente, il suo lavoro mira all’identificazione di nuovi target molecolari e potenziali terapie per l’Atassia di Friedreich, attraverso approcci di “drug discovery” e “drug repurposing”. I programmi di drug discovery nascono da ipotesi originali e dalla scoperta del coinvolgimento del pathway ubiquitina-proteasoma nella degradazione della frataxina, la proteina difettiva nell’Atassia di Friedreich, e hanno portato all’identificazione della E3 ligasi responsabile della degradazione della frataxina, come nuovo target terapeutico per l’Atassia di Friedreich. I programmi di durg repurposing, basati sull’idea di trovare una nuova indicazione terapeutica per un farmaco già approvato, hanno portato all’identificazione del farmaco anti-retrovirale etravirina come promettente candidato per la cura dell’Atassia di Friedreich. Un trial clinico di fase II è attualmente in corso per testare la sicurezza e l’efficacia di etravirina su pazienti affetti da Atassia di Friedreich. Entrambi i programmi di ricerca hanno portato alla creazione di brevetti e pubblicazioni, dei quali la Dott.ssa Rufini è inventore e autore.
Dal 2019 è professore a contratto di Immunology and Immunopathology (Med/04) del corso di laurea in Medicine and Surgery, e di General Pathology (Med/04) del corso di laurea in Dentistry and Dental Prosthetics, presso la Saint Camillus International University of Health and Medical Sciences (UniCamillus).

SCIENTIFIC ACTIVITY
Dr Alessandra Rufini performs her research in the Laboratory of Signal Transduction (www.labst.org), Rare Disease Unit, in the Biomedicine and Prevention Department, at the University of Rome “Tor Vergata”. During her PhD, she focused her studies on the cross-talk between caspases and tyrosine-kinases in the control of the apoptotic pathway. For the last 15 years her main research area has been Friedreich ataxia, a rare neurodegenerative disease that has no cure. Currently, her research is aimed at the identification of new molecular targets and potential therapies for Friedreich ataxia, following both drug discovery and drug repurposing strategies. The drug discovery programs build on original hypothesis and the discovery of the involvement of the ubiquitin-proteasome system in the degradation of frataxin, the protein defective in Friedreich ataxia, and led to the identification of the E3 ligase responsible for frataxin degradation as a new therapeutic target for Friedreich ataxia. The drug repurposing programs, through the screening of an FDA-approved drugs collection, led to the identification of the anti-retroviral drug etravirine as a promising drug candidate for Friedreich ataxia. A phase II clinical trial to test the efficacy of etravirine in a cohort of FRDA patients is currently ongoing. Both research programs led to the creations of patents and publications, of which Dr. Rufini is inventor and author.
Since 2019, Dr Rufini is Contract Professor of Immunology and Immunopathology (Med/04) in the Medicine and Surgery Degree and of General Pathology (Med/04) in the Dentistry and Dental Prosthetics Degree at the Saint Camillus International University of Health and Medical Sciences (UniCamillus).
